Hmmm....I still think 60 is a lot of fish, especially for a highly endangered species like this (if in fact it is a new species). So you're saying 30 fish is enough for each technique. The question is..why do you need two techniques to siamensisdb_answer one question? I mean, if one technique is enough to give you a conclusive set of dat...then there is no need to use a second technique and harm additional 30 fish. *In fact, you can use the same DNA samples (from the first 30 fish) in your second technique.* This will not compromise your experimental design or data analysis because the two techniques are independent from each other (and you will analyze those data separately anyway -- so you won't be comparing samples from the same fish). Please don't take this personally, I'm just trying to help you figure out the way to do your research more efficiently. Since you mentioned shortage in material, this may help you solve that problem.  

On a seperate note, I have a general question concerning the way this research proposal was approved. This kind of research, which involved invasive procedure on animal subjects, needed to be reviewed and supervised by some sort of "Institutional Animal Care Unit or Committee" to ensure proper treatment of animals and to make sure that not too many animals are unnecesssarily harmed. I'm surprised they haven't addressed the isssue of "number of animals" during that review process. I'm even curious if there was such a review process at all. If not, it is something that has to be initiated in your institution!

Please understand that I'm not protesting your research in any way. I agree it is important to conduct this kind of research. But I also want to make sure we don't jeopardize the existence of a potential new species.

Thank u very much about ur suggestion. Im the one person that care about our nat. resource especially fishes.But in this 2 techniques is sapparate by 2 person who have response. Now in my lab. have 3 reserchers that study about Betta. Im first. the sample size for each technique was assigned by our advisor. But I know well about sittuation of this fishes and agree with your conclusion. I will discuss with my team to reduce sample size. But in ur separate note. I dont know about "Institutional Animal Care Unit or Committee" and my advisor dont tell me about it. U have many knowledge about fish research. Please take respect from me....

Thank you for your quick response krub. And thank you for agreeing to discuss the research plan with your team. I wish you best luck and success.

I understand that the two students have their own responsibility. I do hope that they can collaborate (helping each other extracting DNA) and share research material (DNA samples). The data they obtain will, in fact, confirm each other's result (because they use DNA from the same fish). This way, they can graduate quicker, with their own independent works, and use fewer fish.

I think the best way to make sure that you use specimens which really are wild types, that resemble those that were originally described, is to have at least two independent fish taxonomy experts identify each specimen based on the information (e.g. dichotomous keys and/or despriptions) from the original publications. For example, you would need to base the identification of Betta splendens with the description published by Raegan in 1910.
